Specific Inhibition of Chloroplast Replication in Euglena Gracilis by Nalidixic Acid

In Euglena gracilis the synthesis of the chloroplast from its precursor, the proplastid, is separated in time from the division or replication of the organelle which occurs just prior to cell division (10, 12). The proplastids are only found in darkgrown cells and rapidly form chloroplasts when such cells are exposed to light. The proplastids divide during dark growth. Ultraviolet irradiation i...

Influence of growth conditions upon the number of chloroplast DNA molecules in Euglena gracilis.

The number of chloroplast DNA molecules in Euglena gracilis cells was measured by determining the shift in the observed second-order rate constant for the reassociation of (125)I-labeled chloroplast DNA in the presence of unlabeled total cell DNA. Cells grown to stationary phase in the dark contained 217 molecules of chloroplast DNA. Cells grown to stationary phase in the light in either hetero...

Evolution of the chloroplast genome in photosynthetic euglenoids: a comparison of Eutreptia viridis and Euglena gracilis (Euglenophyta).

The chloroplast genome of Eutreptia viridis Perty, a basal taxon in the photosynthetic euglenoid lineage, was sequenced and compared with that of Euglena gracilis Ehrenberg, a crown species. Several common gene clusters were identified and gene order, conservation, and sequence similarity was assessed through comparisons with Euglena gracilis. Significant gene rearrangements were present betwee...

Transcription program of the chloroplast genome of Euglena gracilis during chloroplast development.

RNA transcription in Euglena gracilis chloroplasts has been characterized by hybridization of RNA from cells at different stages of chloroplast development to (3)H-labeled chloroplast DNA restriction endonuclease fragments. Chloroplast DNA was digested into five fragments of 53,000, 35,000, 25,000, 10,000, and 6900 base pairs with Pst I. The 53,000-base-pair DNA was further cleaved by BamHI dig...

Diversity of the ribosomal structures in the Euglena gracilis chloroplast genome: description of a mutant with two ribosomal operons and possible mechanism for its production.

We studied the bleached mutant of Euglena gracilis Y3BUD producing carotenoids and rich in chloroplast DNA. This mutant which was produced more than twenty years ago segregates permanently, forming 30 to 50% daughter cells without carotenoids and poor in chloroplast DNA. Electron microscopy and mapping with restriction enzymes show the deletion of a complete ribosomal operon out of the three pr...
